If you're asking how to remove the instance that you created by clicking Run Now, there's no direct way to do that. I suppose if you deleted the schedule itself and recreated, then that would effectively make the instance not run 12 hours later because the schedule wouldn't exist anymore and thus Blue Prism would have no tasks to run. The other way would be to manipulate the database records directly which isn't encouraged.
The suggested way to get around that is to have access to a remote machine that is set to the same time zone as the server so that the Run Now feature works as expected. I suppose another way is to, instead of using Run Now, you probably could set the schedule to run once and set the datetime to 12 hours in the past, and when you save it, that should effectively do the same thing as Run Now. Fun times.

That issue has been annoying for quite a while. Blue Prism is definitely aware of it, and I believe they attempted a fix in 6.7/6.8 or so but then I think they reverted it. Anyway, they'll prolly fix that eventually.
